The use of cannabidiol (CBD) in dogs with cancer has garnered significant attention in recent years due to its potential therapeutic benefits. CBD, a non-psychoactive compound derived from the hemp plant, interacts with the endocannabinoid system to provide various health benefits without the intoxicating effects associated with tetrahydrocannabinol (THC). 

Emerging research suggests that CBD may offer direct anti-tumor properties, reduce cancer cell proliferation, and induce apoptosis in canine cancer cells. Additionally, CBD's anti-inflammatory and analgesic effects can alleviate pain and improve the overall quality of life for dogs undergoing cancer treatment. As interest in integrative and supportive cancer therapies grows, CBD is increasingly recognized as a promising adjunctive treatment for canine cancer patients.

The endocannabinoid system (ECS) is a complex cell-signaling system identified in the early 1990s by researchers exploring THC, a well-known cannabinoid. This system is active in the body of most mammals, including dogs and humans. 

The endocannabinoid system has three components: endocannabinoids, receptors, and enzymes. The endocannabinoids include Anandamide (AEA) and 2-Arachidonoylglycerol (2-AG). AEA is called the "bliss molecule" because it regulates mood, memory, appetite, and pain sensation. 2-Arachidonoylglycerol (2-AG), found in higher concentrations in the brain, plays a role in regulating the circulatory system, immune response, and neuroprotection.

Known receptors within the endocannabinoid system include CB1 Receptors and CB2 Receptors. Primarily located in the brain and central nervous system, CB1 receptors influence functions such as pain perception, mood, and appetite. Predominantly found in the peripheral organs, especially cells associated with the immune system, CB2 receptors help modulate inflammation and immune response.

The enzyme Fatty Acid Amide Hydrolase (FAAH) breaks down anandamide after it is used within the body. Monoacylglycerol Lipase (MAGL) breaks down 2-AG after it is used. 

The ECS helps maintain homeostasis, ensuring the stability of the body's internal environment. It plays a pivotal role in regulating a variety of physiological and cognitive processes, including the modulation of pain perception, influences of mood regulation and emotional responses, hunger control, digestive processes, regulate the immune system activity and inflammatory response, protects neurons, and aids in the development of neural pathways.

CBD (cannabidiol) products come in three main types: full spectrum, broad spectrum, and isolate. Each type varies in its cannabinoid content, potential effects, and applications.

Full-spectrum CBD contains all cannabinoids found in the cannabis plant, including CBD, THC (tetrahydrocannabinol), terpenes, flavonoids, and other plant compounds. This combination produces an "entourage effect," where the cannabinoids and other compounds work synergistically to enhance therapeutic benefits. 

Although full-spectrum CBD contains trace amounts of THC (less than 0.3%), it is not enough to cause psychoactive effects but can contribute to the entourage effect. It is often used for comprehensive health benefits, including pain relief, anxiety reduction, and anti-inflammatory effects. For example, full-spectrum CBD has shown efficacy in reducing breast cancer cell viability and expression of growth factor receptors.

Broad-spectrum CBD contains multiple cannabinoids and terpenes found in the cannabis plant but has all THC removed. It still benefits from the entourage effect without the risk of THC exposure, making it completely THC-free. Broad-spectrum CBD is suitable for individuals who want to avoid THC for legal, personal, or employment reasons while still benefiting from multiple cannabinoids. For example, broad-spectrum CBD has been reported to be more effective in stress management compared to CBD isolate.

CBD isolate is pure without other cannabinoids, terpenes, or plant compounds. It does not benefit from the entourage effect but provides a high concentration of CBD and is entirely THC-free. CBD isolate is ideal for individuals who need a high dose of CBD or those who are sensitive to other cannabinoids and terpenes. For example, CBD isolate is often used in clinical settings and has been part of studies assessing its efficacy in treating conditions like epilepsy.

The primary differences between full spectrum, broad spectrum, and isolate CBD preparations lie in their cannabinoid content and potential effects. Full-spectrum CBD offers the widest range of benefits through the entourage effect, broad-spectrum CBD provides benefits without THC, and CBD isolate offers pure CBD without other cannabinoids. 

Choosing the right type depends on individual health needs and preferences. Dog parents should request a certificate of analysis that ensures what is in the product when it is produced. The report provides the specific quantity of each cannabinoid, like CBD and THC, that is in the product. A third-party, independent laboratory should generate the report to ensure a non-biased report.

The Use of CBD for Managing Cancer in Dogs

CBD has demonstrated the potential to help manage clinical signs and side effects associated with cancer and its treatment, such as pain, anxiety, nausea, and inflammation. Studies also suggest CBD has significantly reduced cancer cell proliferation in canine neoplastic cell lines.

CBD has been tested both alone and in combination with standard chemotherapeutics like vincristine and doxorubicin. Studies suggest that CBD might reduce cell proliferation in a synergistic or additive manner when combined with these drugs, though the results with doxorubicin are less clear. 

The effects of CBD on cell signaling pathways indicate its role in inducing autophagy and apoptosis, which are processes involved in cell death and cancer treatment. CBD has been observed to induce apoptosis and disturb mitochondrial function in both human and canine glioma cells. It exerts these effects through a mechanism involving dysregulation of calcium homeostasis and mitochondrial activity. These findings underscore CBD's potential as a cytotoxic agent against cancer cells, promoting cell death through pathways independent of cannabinoid receptors.

Cannabinoids can modulate multiple cancer-related pathways, including inhibition of cell growth, induction of apoptosis, and inhibition of metastasis through anti-angiogenesis. They act on cannabinoid receptors (CB1, CB2) and other cellular targets. Preclinical studies have shown that cannabinoids can inhibit the growth of various cancer types, such as glioblastoma, breast cancer, and leukemia.

Studies have shown that CBD, along with other cannabinoids like cannabigerol (CBG), can inhibit the growth of skin carcinoma cells. Research involving Haccat human skin carcinoma cells demonstrated that both CBD and CBG, as well as their combination, have significant anti-tumor activity, suggesting their potential as therapeutic agents against skin cancer.

Another anticancer property of CBD is that it has been shown to induce apoptosis (programmed cell death) and inhibit angiogenesis (formation of new blood vessels) in melanoma cells, reducing tumor growth. This effect is mediated through the activation of cannabinoid receptors (CB1 and CB2), which are expressed in both normal skin and skin tumors. Local administration of cannabinoid receptor agonists has been shown to induce significant growth inhibition of malignant tumors in mice, accompanied by increased apoptotic cell death and impaired tumor vascularization.

CBD exhibits anti-angiogenic properties through various physiological mechanisms, including the suppression of vascular endothelial growth factor (VEGF) and hypoxia-inducible factors (HIF-1α), modulation of ERK and Akt pathways, induction of TIMP-1, inhibition of EMV release, and HO-1 mediated effects on endothelial cells. These multi-target effects of CBD contribute to its potential as an anticancer agent by inhibiting the formation of new blood vessels necessary for tumor growth.

The ERK and Akt pathways are central to regulating essential cellular functions. Their dysregulation is common in various cancers, making them critical targets for therapeutic intervention. CBD can modulate cell survival, growth, proliferation, metabolism, angiogenesis, and protein synthesis of the cancer cells by inhibiting these pathways.

Using CBD for Pain Management for Dogs With Cancer

Like every mammal, dogs possess an endocannabinoid system, a network of receptors throughout the body that modulates pain, anxiety, and nausea. By interacting with this system, CBD can mimic the effects of naturally occurring cannabinoids, effectively managing various types of pain and providing significant relief.

CBD has shown promise in addressing arthritis, inflammation, and other discomforts that often accompany cancer in dogs. Its anti-inflammatory properties can alleviate these clinical signs, improving affected dogs' overall quality of life.

Additionally, certain cancers can induce seizures, which are distressing for both the dog and the owner. CBD has been demonstrated to reduce the frequency, duration, and severity of seizures in both humans and dogs. This capability makes CBD a compelling addition to a dog's cancer treatment regimen, particularly for managing seizure-related symptoms.

Pain and inflammation are common side effects of cancer treatments and the disease's progression. Effective pain management is crucial in treating canine cancer. While consulting with a veterinarian is essential, incorporating CBD into the pain management plan can offer an additional layer of relief naturally and with minimal side effects. This holistic approach can enhance the well-being of dogs undergoing cancer treatment, providing comfort and improving their quality of life.

The endogenous cannabinoid system, including cannabinoid receptors and endocannabinoids, regulates tumor growth and immune response. Exogenous cannabinoids like THC have been shown to inhibit tumor growth in a receptor-dependent manner, suggesting that targeting the endocannabinoid system could be a viable strategy for cancer treatment. CBD shows promise as a therapeutic agent for cancer, capable of inhibiting tumor growth, inducing apoptosis, and enhancing the effects of conventional cancer treatments. 

CBD has shown significant promise in managing clinical signs and side effects associated with cancer in dogs, such as pain, anxiety, nausea, and inflammation.

Emerging research indicates that CBD may possess direct anticancer properties, including reducing cancer cell proliferation, inducing apoptosis, and inhibiting metastasis through anti-angiogenesis.

CBD is effective in managing the frequency, duration, and severity of seizures in dogs with cancer.
